LentiCRISPR and CRISPR/Cas 9 mediated enhancer deletion using puromycin selection vector to produce useful reagents in apoptosis research studies
I got the opportunity to intern in the Host-Pathogen Interaction and Signaling lab in Presidency University, Kolkata. I worked under the guidance of Dr. Debanjan Mukhopadhyay. My short-term project focused on designing sgRNAs for CASP7, NOX1, IRF, JAK, STAT, CYLD and cloning them into stbl3 E. coli using pX459 V2.0 plasmid vector through CRISPR genome engineering. The larger aim is to transfect U-20S osteosarcoma cells with the engineered plasmid to knock out those specific genes, to study the apoptosis and JAK-STAT pathway. LentiCRISPR method was used for the genes GBP1, GBP3, GBP4, GBP6.
